The slow development of cost-effective medical microdevices with strong analytical performance characteristics is due to a lack of selective and efficient analyte capture and signaling. The recently developed programmable bio-nano-chip (PBNC) is a flexible detection device with analytical behavior rivaling established macroscopic methods. The PBNC system employs ≈300 μm-diameter bead sensors composed of agarose “nanonets” that populate a microelectromechanical support structure with integrated microfluidic elements. The beads are an efficient and selective protein-capture medium suitable for the analysis of complex fluid samples. Microscopy and computational studies probe the 3D interior of the beads. The relative contributions that the capture and detection of moieties, analyte size, and bead porosity make to signal distribution and intensity are reported. Agarose pore sizes ranging from 45 to 620 nm are examined and those near 140 nm provide optimal transport characteristics for rapid (<15 min) tests. The system exhibits efficient (99.5%) detection of bead-bound analyte along with low (≈2%) nonspecific immobilization of the detection probe for carcinoembryonic antigen assay. Furthermore, the role analyte dimensions play in signal distribution is explored, and enhanced methods for assay building that consider the unique features of biomarker size are offered.

A new generation of programmable diagnostic devices is needed to take advantage of information generated from the study of genomics, proteomics, metabolomics and glycomics. This report describes the ‘programmable nano-bio-chip’ with potential to bridge the significant scientific, technology and clinical gaps through the creation of a diagnostic platform to measure the molecules of life. This approach, with results at the point-of-care, possesses capabilities for measuring such diverse analyte classes as cells, proteins, DNA and small molecules in the same compact device. Applications such as disease diagnosis and prognosis for areas including cancer, heart disease and HIV are described. New diagnostic panels are inserted as ‘plug and play’ elements into the modular platform with universal assay operating systems and standard read out sequences. The nano-bio-chip ensemble exhibits excellent analytical performance and cost−effectiveness with extensive validation versus standard reference methods (R2 = 0.95–0.99). This report describes the construction and use of two major classes of nano-bio-chip designs that serve as cellular and chemical processing units, and provides perspective on future growth in this newly emerging field of programmable nano-bio-chip sensor systems.

The development of a micromachined fluidic structure for the introduction of liquid samples into a chip-based sensor array composed of individually addressable polymeric microbeads is presented. The micromachined structure consists of micromachined storage cavities combined with a covering glass layer that confines the microbeads and fluidic channels. In our sensor array transduction occurs via optical (colorimetric and fluorescence) changes to receptors and indicator molecules that are covalently attached to termination sites on the polymeric microbeads. Spectral data are acquired for each of the individual microbeads using a charged-coupled device (CCD) allowing for the near-real-time analysis of liquid sample. Hence the micromachined fluidic structure must allow for both optical access to the microbeads and fluid flow through the micromachined cavities that serve as the microreactors/analysis chambers. One of the key parts of the structure is a passive fluid introduction system driven only by capillary force. This simple means of fluid introduction realizes a compact device. The capillary flow on the inlet channel has been studied, and the responses of the microbeads (alizarin complexone) to a liquid sample have been characterized. The test results show that this system is useful in a micro-total-analysis-system (μ-TAS) and biomedical applications.

n the last decade, saliva has been advocated as a non-invasive alternative to blood as a diagnostic fluid. However, use of saliva has been hindered by the inadequate sensitivity of current methods to detect the lower salivary concentrations of many constituents compared to serum. Furthermore, developments in the areas related to lab-on-a-chip systems for saliva-based point of care diagnostics are complicated by the high viscosity and heterogeneous properties associated with this diagnostic fluid. The biomarker C-reactive protein (CRP) is an acute phase reactant and a well-accepted indicator of inflammation. Numerous clinical studies have established elevated serum CRP as a strong, independent risk factor for the development of cardiovascular disease (CVD).

Here we describe the development of a prototype for a simple, rapid, and affordable method for counting CD4 lymphocytes. Microliter volumes of blood without further sample preparation are stained with fluorescent antibodies, captured on a membrane within a miniaturized flow cell and imaged through microscope optics with the type of charge-coupled device developed for digital camera technology. An associated computer algorithm converts the raw digital image into absolute CD4 counts and CD4 percentages in real time. The accuracy of this prototype system was validated through testing in the United States and Botswana, and showed close agreement with standard flow cytometry ( r = 0.95) over a range of absolute CD4 counts, and the ability to discriminate clinically relevant CD4 count thresholds with high sensitivity and specificity.

Nanometer-size threads of amphiphilic polythiophene are demonstrated to form upon collapsing a monolayer of polythiophene on a Langmuir–Blodgett trough (see also the cover). Bundles of these nanowires can be manipulated to fill the gap between gold electrodes, as shown in the Figure, where the wires are depicted in dark gray and the gold electrodes in light gray.

Methods capable of forming highly organized monolayers on top of YBa 2 Cu 3 O 7 (YBCO), a high- T c superconductor, have been identified and are described for the first time. Here, grazing reflectance infrared fourier transform spectroscopy (GRIFTS) is employed to evaluate the degree of order for these monolayer structures. Through these investigations, it is found that while octadecylamine forms a well-ordered, crystalline-like monolayer on the surface of c -axis-oriented YBa 2 Cu 3 O 7 thin films, the same reagent adsorbed onto polycrystalline YBa 2 Cu 3 O 7 pellets affords disordered, liquid-like monolayers...

Electrochemical techniques are exploited to fabricate conductive polymer/high- Tc superconductor bilayer structures. Scanning electron microscopy, atomic force microscopy, and electrochemical techniques are utilized to characterize the electrodeposition of polypyrrole layers grown onto YBa~Cu3076 films. In such hybrid polymer/superconductor systems, it is found that, when the polymer is oxidized to its conductive state, the transition temperature (Tc) and critical currents (J,) of the underlying superconductor film are suppressed. Reversible modulations of the transition temperatures of up to 50 K are noted for these structures. Upon reduction of the conductive polymer layer back to its nonconductive form. both T, and J, are found to return to values close to those acquired for the underivatized YBa2Cu3076 film.
